Gene targeting strategy for B-hCFD mice.
The exons 1-5 of mouse Cfd gene that encode the full-length protein were replaced by human CFD exons 1-5 in B-hCFD mice.
Strain specific analysis of CFD gene expression in wild-type C57BL/6 mice and homozygous B-hCFD mice by RT-PCR. Mouse Cfd mRNA was detectable in heart tissue of wild-type C57BL/6 mice (+/+). Human CFD mRNA was detectable only in homozygous B-hCFD mice (H/H) but not in wild-type mice.
Strain specific CFD expression analysis in wild-type C57BL/6 mice and homozygous B-hCFD mice by western blot. Fat tissue was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCFD mice (H/H). Human monocytic leukemia cells (THP-1) were used as positive control.
