Targeting strategy
Gene targeting strategy for B-hDKK1 mice. The exons 1-4 of mouse Dkk1 gene that encode the full-length protein was replaced by human DKK1 exons 1-4 in B-hDKK1 mice.
mRNA expression analysis
Strain specific analysis of DKK1 gene expression in wild-type C57BL/6 mice and homozygous B-hDKK1 mice by RT-PCR. Mouse Dkk1 mRNA was detectable in thymus of wild-type C57BL/6 mice (+/+). Human DKK1 mRNA was detectable only in homozygous B-hDKK1 mice (H/H) but not in wild-type mice.
Protein expression analysis
Strain specific DKK1 expression analysis in wild-type C57BL/6 mice and homozygous B-hDKK1 mice by ELISA. Serum was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hDKK1 mice (H/H). Mouse DKK1 was only detectable in wild-type mice. DKK1 was both detectable in wild-type C57BL/6 mice and homozygous B-hDKK1 mice. Therefore, it is speculated that this anti-human DKK1 antibody is cross-reactive between human and mouse.
