RT-PCR analysis of IL12A and IL12B gene. The human IL12A but not mouse Il12a mRNA was detectable in splenocytes of the homozygous B-hIL12A/hIL12B mice. The human IL12B but not mouse Il12b mRNA was detectable in thymocytes of the homozygous B-hIL12A/hIL12B mice.
Strain specific IL12(heterodimer p70) expression analysis in homozygous B-hIL12A/hIL12B mice by ELISA. Serum was collected om WT (+/+) and homozygous B-hIL12A/IL12B mice (H/H) mice stimulated with anti-mCD3ε and anti-mCD28 in vivo (n=3), and analyzed by ELISA with species-specific IL12 ELISA kit. Mouse IL12 (p70) was detectable in WT mice. Human IL12 (p70) was exclusively detectable in homozygous B-hIL12A/IL12B mice (H/H) mice, but not in WT mice. ND: Not detectable.
