Western blot analysis of JAG1 protein expression in homozygous B-hJAG1 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hJAG1 mice (H/H), and then analyzed by western blot with anti-JAG1 antibody. 72 μg total proteins were loaded for western blotting analysis. JAG1 protein was detected in the kidney, lung, heart, spleen of wild-type C57BL/6 mice and homozygous B-hJAG1 mice, as the antibody is cross-recognize both human and mouse JAG1.
Strain specific analysis of JAG1 mRNA expression in wild-type C57BL/6 mice and B-hJAG1 mice by RT-PCR. Lung RNA were isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hJAG1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human JAG1 primers. Mouse Jag1 mRNA was detected in wild-type C57BL/6 mice. Human JAG1 mRNA was detected in homozygous B-hJAG1 mice but not in wild-type mice.
