Gene targeting strategy for B-hRGMA mice. The exon 3 and exon 4 of mouse Rgma gene was replaced by human RGMA exon 3 and exon 4 in B-hRGMA mice.
Strain specific analysis of RGMA gene expression in wild-type C57BL/6 mice and hRGMA mice by RT-PCR. Mouse Rgma mRNA was detectable only in the wild-type C57BL/6 mice (+/+). Human RGMA mRNA was detectable only in homozygous B-hRGMA mice (H/H), but not in wild-type mice.
Strain specific RGMA expression analysis in homozygous B-hRGMA mice by western blot. Brain tissue was collected from the wild-type mice (+/+) and homozygous B-hRGMA (H/H) mice, and analyzed by western blot with anti-RGMA antibody. RGMA was detectable in wild-type mice (+/+) and homozygous B-hRGMA mice (H/H) due to the cross-reactivity of antibodies.
