      • SIGLEC6 is an inhibitory receptor selectively expressed on human mast cells and is a member of a family of receptors that bind to sialoglycan.
      • The full coding sequences of human SIGLEC6 gene, including the promoter, 5’UTR and 3’UTR are inserted into mouse Hipp11 (H11) locus in B-hSIGLEC6 mice.
      • Human SIGLEC6 mRNA was exclusively detectable in homozygous B-hSIGLEC6 mice.
      • Human SIGLEC6 protein was detectable on mast cells, B cells, NK cells, DCs, neutrophils, monocytes and macrophages of homozygous B-hSIGLEC6 mice.

      mRNA expression analysis

      Species specific analysis of SIGLEC6 gene expression in wild-type C57BL/6JNidfc mice and homozygous humanized B-hSIGLEC6 mice by RT-PCR. Spleen was collected from wild-type C57BL/6JNidfc mice (+/+) and homozygous B-hSIGLEC6 mice (H/H). Human SIGLEC6 mRNA was detectable only in homozygous B-hSIGLEC6 mice, but not in wild-type C57BL/6JNidfc mice.

      Protein expression analysis in mast cells

      Strain specific SIGLEC6 expression analysis in homozygous B-hSIGLEC6 mice by flow cytometry. Peritoneal lavage fluid was collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hSIGLEC6 mice (H/H), and protein expression was analyzed with anti-human SIGLEC6 antibody (BD, 747915) by flow cytometry. hSIGLEC6 was only detectable in mast cells of homozygous B-hSIGLEC6 mice but not in wild-type C57BL/6JNifdc mice.

      Protein expression analysis in T, B and NK cells

      Strain specific SIGLEC6 expression analysis in homozygous B-hSIGLEC6 mice by flow cytometry. Splenocytes, blood, and lymph nodes were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hSIGLEC6 mice (H/H), and protein expression was analyzed with anti-human SIGLEC6 antibody (BD, 747915) by flow cytometry. hSIGLEC6 was detectable in B and NK cells in the spleen and lymph nodes of homozygous B-hSIGLEC6 mice, and hSIGLEC6 was detectable in T, B, and NK cells in the blood of homozygous B-hSIGLEC6 mice.
