        Gene targeting strategy for B-hSLC16A3 mice. The exons 2-4 of mouse Slc16a3 gene that encode the full-length protein were replaced by human SLC16A3 exons 2-5 in B-hSLC16A3 mice.

        Protein expression analysis
        Western blot analysis of SLC16A3 protein expression in homozygous B-hSLC16A3 mice. Various tissue lysates were collected from wild-type C57BL/6N mice (+/+) and homozygous B-hSLC16A3 mice (H/H), and then analyzed by simple-western blot with anti-SLC16A3 antibody. 3 μg total proteins were loaded for western blotting analysis. SLC16A3 protein was detected in the brain，heart，liver，lung and skeletal muscle of wild-type C57BL/6N mice and homozygous B-hSLC16A3 mice, as the antibody is cross-recognize both human and mouse SLC16A3.
        mRNA expression analysis

        Strain specific analysis of SLC16A3 mRNA expression in wild-type C57BL/6N mice and B-hSLC16A3 mice by RT-PCR. Skeletal muscle RNA were isolated from wild-type C57BL/6N mice (+/+) and homozygous B-hSLC16A3 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human SLC16A3 primers. Mouse Slc16a3 mRNA was detectable only in wild-type C57BL/6N mice. Human SLC16A3 mRNA was detectable only in homozygous B-hSLC16A3 mice but not in wild-type mice.
