Gene targeting strategy for B-hTFPI mice. The exons 3-9 of mouse Tfpi gene that encode the full-length protein were replaced by human TFPI exons 2-8 in B-hTFPI mice.
Strain specific TFPI expression analysis in wild type (WT) mice and B-hTFPI mice by ELISA. Plasma were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTFPI mice (H/H), and analyzed by ELISA with TFPI ELISA kit. Mouse TFPI was exclusively detectable in wild-type mice. TFPI was detectable in homozygous B-hTFPI mice (H/H) and wild-type mice (+/+) due to the cross-reactivity of antibodies. ND: not determined.
