Gene targeting strategy for B-hTGM2 mice. The exons 1-13 of mouse Tgm2 gene that encode the full coding sequences were replaced by human TGM2 exons 1-13 in B-hTGM2 mice.
Strain specific analysis of TGM2 gene expression in wild-type C57BL/6 mice and B-hTGM2 mice by RT-PCR. Mouse Tgm2 mRNA was detectable in ovary of wild-type C57BL/6 mice (+/+) but not in homozygous B-hTGM2 mice (H/H). Human TGM2 mRNA was detectable only in homozygous B-hTGM2 mice but not in wild-type mice.
Strain specific TGM2 expression analysis in homozygous B-hTGM2 mice by flow cytometry. Lung and spleen tissues were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTGM2 mice (H/H) and analyzed by western blotting with anti-TGM2 antibody. Hela cell line was used as a positive control for human TGM2 protein. TGM2 was detectable in wild-type mice and homozygous B-hTGM2 mice with a cross-reactive anti-TGM2 antibody.
