• Background: Complement component C3 is a 190 kDa protein that is the most prevalent complement protein in serum and is involved in the production of fragments C3a and C3b, which are crucial for complement activation and immune response. It is also necessary for all three complement-activating pathways: classical, lectin, and alternative.
• Gene targeting strategy: The exons 2-40 of mouse C3 gene were knocked out in B-C3 KO mice. As a result, mouse C3 protein not expressed anymore.
• Validation: Mouse C3 was only detectable in wild-type C57BL/6JNifdc mice but not in B-C3 KO mice.

Strain specific mouse C3 expression analysis in wild-type C57BL/6JNifdc mice and B-C3 KO mice by ELISA. Serum was collected from wild-type C57BL/6JNifdc mice (+/+) (male, n=3, 8-week-old) and B-C3 KO mice (-/-) (male, n=3, 8-week-old). The expression level of mouse C3 were analyzed by ELISA (mouse C3 ELISA kit: Abcam, ab157711). Mouse C3 was only detectable in wild-type C57BL/6JNifdc mice but not in B-C3 KO mice.
