Gene targeting strategy for B-hIL2RG mice. The exons 1~8 of mouse Il2rg gene that encode the full-length protein were replaced by human IL2RG exons 1~8 in B-hIL2RG mice.
Strain specific IL2RG expression analysis in hemizygous B-hIL2RG mice by flow cytometry.
Splenocytes were collected from wild-type C57BL/6 mice (+/+) and hemizygous B-hIL2RG mice (H/Y), and analyzed by flow cytometry with species-specific anti-IL2RG antibody. Mouse IL2RG was detectable in T cells of wild-type mice. Human IL2RG was exclusively detectable in T cells of hemizygous B-hIL2RG mice but not wild-type mice.
