Gene targeting strategy for B-hCCR4 mice. The exon 2 of mouse Ccr4 gene that encodes the full-length protein was replaced by human CCR4 exon 2 in B-hCCR4 mice.
Strain specific analysis of CCR4 gene expression in WT and hCCR4 mice by RT-PCR. Mouse Ccr4 mRNA was detectable only in splenocytes of wild-type (+/+) mice. Human CCR4 mRNA was detectable only in H/H, but not in +/+ mice.
Strain specific CCR4 expression analysis in homozygous B-hCCR4 mice by flow cytometry. Splenocytes were collected from WT and homozygous B-hCCR4 (H/H) mice, and analyzed by flow cytometry with species-specific anti-CCR4 antibody. Mouse CCR4 was detectable in WT mice. Human CCR4 was exclusively detectable in homozygous B-hCCR4 but not WT mice.
