Gene targeting strategy for B-hCD209 mice.
The exons 3-7 of mouse Cd209a gene that encode the extracellular domain were replaced by human CD209 exons 3-7 in B-hCD209 mice.
Strain specific analysis of CD209 mRNA expression in wild-type C57BL/6 mice and B-hCD209 mice by RT-PCR. Spleen RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCD209 mice (H/H), then cDNA libraries were synthesized by reverse transcription, and followed by PCR with mouse or human CD209 primers. Mouse CD209a mRNA was detectable only in wild-type C57BL/6 mice. Human CD209 mRNA was detectable only in homozygous B-hCD209 mice but not in wild-type mice.
