Gene targeting strategy for B-hITGB2 mice. The exons 3-15 of mouse Itgb2 gene that encode extracellular domain is replaced by human counterparts in B-hITGB2 mice. The genomic region of mouse Itgb2 gene that encodes transmembrane domain and cytoplasmic portion is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained. The chimeric ITGB2 expression is driven by endogenous mouse Itgb2 promoter, while mouse Itgb2 gene transcription and translation will be disrupted.
Strain specific CD18/ITGB2 expression analysis in homozygous B-hITGB2 mice by flow cytometry. Splenocytes and blood was collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hITGB2 mice (H/H), and analyzed by flow cytometry with species-specific anti-CD18 antibody (Biolegend, 373405; Biolegend, 101408). Mouse CD18 was detectable in wild-type mice. Human CD18 was exclusively detectable in homozygous B-hITGB2 mice but not in wild-type mice.
