Gene targeting strategy for B-hPSCA mice. The full coding region of mouse Psca gene was replaced by human PSCA counterpart gene sequences in B-hPSCA mice.
Strain specific analysis of PSCA gene expression in wild-type mice and B-hPSCA mice by RT-PCR. Mouse Psca mRNA was detectable only in stomach of wild-type mice (+/+). Human PSCA mRNA was detectable only in homozygous B-hPSCA mice (H/H) but not in wild-type C57BL/6 mice.
PSCA protein expression analysis in wild-type mice and B-hPSCA mice by western blotting. PSCA was both detectable in B-hPSCA mice and wild-type mice because the antibody has cross reaction between human and mouse. But, it has been confirmed the PSCA sequence was humanization successfully.
